Alpha-1d Adrenoceptor Antagonist Inhibits Premicturition Contractions in Rats with Bladder Outlet Obstruction

چکیده

Aims of Study Lower urinary tract symptoms associated with bladder outlet obstruction include storage and voiding symptoms, and the most bothersome in men with benign prostatic hyperplasia (BPH) is predominantly storage rather than voiding symptoms. Detrusor overactivity, a major cause of irritative symptoms is a common phenomenon in those with BPH. However, the pathophysiology of this detrusor dysfunction remains to be elucidated. Now alpha 1 adrenoceptor (AR1) antagonists are widely used to treat LUTS suggestive of BPH, and this therapeutic agent relieves voiding as well as storage symptoms. The relaxtant effect of AR1 antagonists on the smooth muscle tone in prostatic tissue has been well documented, and this effect is predominantly mediated by AR1a. Because a highly selective AR1a antagonist did not appear to relieve irritative symptoms in patients with BPH despite of a significant increase in urinary flow rate, the role of another distinct subtype of adrenoceptor, especially AR1d adrenoceptor, in the development of storage symptoms has been focused on. Aims of this study was to clarify the effects of a selective AR1d antagonist, naftopidil, on the micturition reflex in rats with bladder outlet obstruction.

An alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate glutamate-receptor antagonist can inhibit premicturition contractions in rats with bladder outlet obstruction.

OBJECTIVE To explore the possible involvement of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) glutamate-receptors in bladder dysfunction associated with bladder outlet obstruction (BOO), as detrusor overactivity (DO) is common in men with benign prostatic hyperplasia. MATERIALS AND METHODS Proposed mechanisms of DO include the myogenic or neurogenic theory, and the autonomous b...

Effects of tamsulosin on bladder blood flow and bladder function in rats with bladder outlet obstruction.

OBJECTIVES To investigate the mechanism underlying the ameliorating effect of tamsulosin, an alpha(1)-adrenoceptor antagonist, on storage symptoms associated with benign prostatic hyperplasia, the effects of tamsulosin on bladder blood flow (BBF) and bladder function was evaluated in rats with bladder outlet obstruction (BOO). METHODS BOO was produced by partial ligature of the proximal ureth...

Synergic Supressive Effect of the Combination Therapy with Silodosin and Imidafenacin on Non-voiding Contractions in Male Rats with Sub- Acute Bladder Outlet Obstruction

Hypothesis / aims of study Currently, the α1-adrenoceptor (α1-AR) antagonist/antimuscarinic combination appears to be a second-line add-on for patients with insufficient symptom relief after monotherapy in males with LUTS associated with bladder outlet obstruction (BOO) (1). However, no urodynamic evaluation has been reported on the effect of the combination therapy. In this study, first, by us...

Key Words: α1A adrenoceptor antagonist, Urethral perfusion pressure, Tamsulosin

Objectives: This study was performed to assess the effects of α1A adrenoceptor antagonists on urethral perfusion pressure (UPP) and their therapeutic potential for female bladder outlet obstruction (BOO). Methods: A cannula was inserted into the femoral arteries of female rats to administer tamsulosin (treatment I), doxazosin (treatment II) or phentolamin (treatment III) and to monitor systemic...

Effects of alpha 1-adrenergic receptor subtype selective antagonists on lower urinary tract function in rats with bladder outlet obstruction.

PURPOSE Antagonists of alpha 1-adrenergic receptors (alpha 1ARs) relieve obstructive and irritative symptoms in patients with bladder outlet obstruction. However, to our knowledge mechanisms underlying the relief of irritative symptoms remain unknown. Because bladder alpha 1dARs are up-regulated in some rats with bladder outlet obstruction, we investigated the effect of the alpha 1aAR antagonis...
